So I booked a Singapore flight through Expedia since it was quite a bit lower than booking through SQ. Because of this I used my CSR instead of Amex Plat. The purchase posted to my account and it posted as a Singapore purchase and not Expedia. Could I have used my Amex Plat and gotten the 5x?

Possibly, but not necessarily. Merchants have different agreements with different card issuers so it can be coded one way with Visa and another with Amex. But yeah if it is actually showing as an SQ charge then you probably could have.
